In 2D gel electrophoresis, proteins are separated based on two different properties: their isoelectric point (pI) and their molecular weight. First, proteins are separated by pI using isoelectric focusing (IEF), which uses a pH gradient to separate proteins by their charge. The resulting protein mixture is then subjected to electrophoresis on a second dimension, where proteins are separated by their molecular weight. The resulting gel can then be visualized and analyzed to identify and quantify individual proteins in the mixture.
